Di2 should last well over 1000 km, especially as lots of the kms on GVBR will be cruising and not changing gears frequently. Take your charger along, but seriously doubt you would need it.

From Essendon the most direct route would be down Mt Alexander Rd, then at Flemington bridge get onto the Capital City trail along the Moonee Ponds Creek, that comes out near Docklands (Costco), then it's bike path through to the city.
